New approaches in pertussis vaccines for developing countries
نویسندگان
چکیده
The currently available pertussis vaccines are highly efficacious. However, whole cell vaccines display some reactogenicity and acellular vaccines are less toxic, but elevated costs limit their use in developing countries. Instituto Butantan, Brasil, has developed a new technology to remove lipopolysaccharides from the whole cell vaccine using a simplified and not expensive methodology, which also provides monophosphoryl lipid A as by-product, to be used as adjuvant. This new whole cell pertussis vaccine with low LPS content combined with diphtheria and tetanus antigens (DTPlow), was shown to be as potent as the regular DTP vaccine and less reactogenic. As another by-product, an alternative acellular pertussis vaccine can be produced by a simple low cost procedure. A recombinant BCG-Pertussis vaccine was developed which can be given at birth as a single dose, potentially covering the neonate susceptibility before immunity conferred by pertussis vaccination schedule becomes effective.

Impact of vaccination on epidemiology of diphtheria and pertussis
Whooping cough and diphtheria are vaccine preventable diseases. Diphtheria, due to Corynebacterium diphtheriae or Corynebactium ulcerans, two Gram positive bacteria, is a serious upper respiratory tract disease with high morbidity and mortality rates. Vaccination, via an acellular vaccine composed only of purified, detoxified diphtheria toxin, has significantly reduced the incidence of the dise...
متن کاملPertussis vaccines: position paper of Indian Academy of Pediatrics (IAP).
Pertussis continues to be a major public health problem in both developing and developed countries. Data on exact burden and incidence of pertussis in the developing countries including India is sparse. However, the disease is widespread, even if not adequately measurable. Pertussis incidence has been increasing steadily in the last decade especially in industrialized countries. Outbreaks are r...
متن کاملPossible options for new pertussis vaccines.
Increasing evidence that the currently available acellular pertussis vaccines are not providing optimal control of pertussis in the United States and many other countries has stimulated interest in improvements of the current vaccines and in the development of new vaccines. A better understanding of the limitations of the current vaccines and the basis for the pertussis resurgence is needed to ...
متن کاملSubcutaneous administration of a fusion protein composed of pertussis toxin and filamentous hemagglutinin from Bordetella pertussis induces mucosal and systemic immune responses
Objective(s): After decades of containment, pertussis disease, caused by Bordetella pertussis seems to be re-emerging and still remains a major cause of reported vaccine-preventable deaths worldwide. The current licensed whole-cell vaccines display reactogenicity while acellular vaccines are expensive and do not induce Th1-type immune responses that are required for optimum protection against t...
متن کاملVirulence Factors Variation Among Bordetella Pertussis Isolates in Iran
Pertussis is still endemic and the recently resurgence of the disease caused by Bordetella pertussis has been shown in many countries. The polymorphism of the virulence genes of B. pertussis and lack of any information about the allelic variation between the Iranian isolates promotes us to analysis of the genes encoded virulence factors including ptxS1, prn, fim3 and cya to understand the diffe...
